UK Regulators Advise on Risk Linked to PPIs

The UK’s Medicines and Healthcare products Regulatory Agency (MHRA) updated its recommendations on the safety of proton pump inhibitors (PPIs)—such as Prilosec, Nexium, Prevacid, and Aciphex - warning that there may be an increased risk of fractures associated with their use.

The agency notes that observational studies on a risk of fractures linked to the drugs suggests a possibility of an increased risk for wrist, spine, or hip fracture, especially if the PPIs are used in high doses over long periods of time. The increased risk was found primarily in elderly patients and it is possible that other factors contribute to the increased risk, according to the agency.

The MHRA also received reports of patients experiencing hypomagnesemia associated with prolonged use of PPIs. Series manifestations of hypomagnesemia include convulsions, fatigue, delirium, dizziness, and ventricular arrhythmia.
