Gateway Analytical to Present Advancements in Particle Sizing Research at Pittcon 2011

Accurate knowledge of the active pharmaceutical ingredient particle size is critical for determining the ultimate dissolution rate in the mucous membrane of the nasal cavity. When preparing dried samples of nasal suspensions for ingredient-specific particle sizing (ISPS) utilizing Raman chemical imaging (RCI), the source of particle aggregation or agglomeration is ambiguous. Particles may exist as true agglomerates, or particles may agglomerate during the drying process.
This study demonstrates the advantages of sample substrate pretreatment for the minimization of particles agglomeration during the sample preparation process. RCI for a nasal spray suspension, along with a polystyrene microsphere sizing standard, will be presented.
Sizing Standard Development for Raman Chemical Imaging

This talk will summarize the progress made in utilizing NIST traceable polystyrene microspheres as a particle sizing calibration standard for wide-field RCI of micron sized Orally Inhaled and Nasal Drug Product particles. Wide-field RCI particle size distributions will be discussed for various morphological parameters. A representative nasal spray sample will be reviewed to demonstrate the application of this methodology.
